Anthropic researcher and Harvard mathematician Levent Alpoge used Claude Fable 5 to produce a counterexample to the Jacobian conjecture — open since 1939. It's a polynomial map from C³ to C³ with constant Jacobian determinant −2 that is not injective: three distinct rational points all map to (−1/4, 0, 0).

The remarkable part is how checkable it is — single-digit integer coefficients, verifiable by hand or Wolfram Alpha in seconds — which is why the math community's reaction has been unusually receptive rather than dismissive. It still awaits a formal arXiv writeup and peer review, but it's the highest-profile open problem an LLM has touched to date.
